The block reward is the newly minted bitcoins that are given to the miner who successfully solves the mathematical puzzle and adds a new block to the blockchain. Currently, the block reward is 6.25 bitcoins. The transaction fees, on the other hand, are the fees paid by users for including their transactions in the block. Miners prioritize transactions with higher fees, so the more transactions and higher fees there are, the higher the mining reward will be. The reward calculation is done automatically by the Bitcoin protocol and is designed to gradually decrease over time as the total supply of bitcoins approaches its maximum limit of 21 million.
